Going by the information provided by some former 727 pilots, all three tanks must have fuel in them. So if you need to 21000 lbs of fuel for a flight, you put 7000 lbs in all three fuel tanks. If you are going to need more than ~36000 lbs of fuel, the extra has to go into the center tank and the above rule applies.
